Lexington has a rush hour like any city, but traffic has gotten bad enough that you'll hit delays even between the usual windows. The best time to get around town is late at night, around 9 or 10pm, when the roads are mostly clear and the only traffic is nightshift workers heading in or evening-shift folks heading home. Night owls have a real advantage here.

During the school year, check whether UK has a home football or basketball game before you head out, since those bring traffic citywide. Without a game on the schedule you'll still hit some traffic, but you're less likely to get stuck somewhere for half an hour.
